The dosage of curcumin to alleviate movement symptoms in a 6-hydroxydopamine-induced Parkinson's disease rat model

BackgroundCurcumin is a natural compound with extensive pharmacological effects. This research to verify the optimal dose and administration duration efficacy of curcumin in alleviating movement symptoms Parkinson's disease (PD).MethodsWistar rats were divided into six groups including control, model, levodopa treatment low/middle/high (40/80/160 mg/kg/d) groups. After stereotactic brain injection 6-hydroxydopamine (6-OHDA), was given by intragastric for 2 weeks. To evaluate drug effect, received behavioral tests apomorphine (APO)-induced rotation test, rotarod test open field test. Then sacrificed slices substantia nigra pars compacta (SNc) used immunofluorescence staining.ResultsAfter 6-OHDA injection, model group showed typical severe APO-induced healthy side, decreased latency constant or accelerative mode, total distance average speed In results staining, induced damage dopaminergic neurons SNc. The 160 mg/kg/d intervene weeks alleviated most disorders but 40/80 limitations. Then, we compared effect 1 week intervention indicated that could better alleviate symptoms.ConclusionsCurcumin 6-OHDA-induced PD rat model. Additionally, against dependent optimally therapeutic effect.
